Current Research Projects
Design and synthesis of novel benzimidazole derivatives with potential biological activities: A number of novel benzimidazole derivatives are being synthesized currently in order to investigate potential biological activities.
Commercial scale synthesis method development for Proton Pump Inhibitor (PPI) raw materials by applying Qbd-based reverse engineering technology: This project is funded by World Bank under conduction of University Grants Commission, Bangladesh. The research project is being currently pursued for the commercial synthesis method development of one of the most commonly used APIs like PPIs (omeprazole, esomeprazole, pantoprazole, rabeprazole) by using Qbd-based reverse engineering technology. Some Qbd methods have been optimized and applied for the synthesis of omeprazole in pilot scale with high yields. The research is being carried out in collaboration with Marshal University, West Virginia, USA.
Research Experience
●Syntheses of N-nonpolar nucleobase dipeptides by Ugi four- component condensation reaction: A novel class of a,a- and a,b-dipeptides having fluoroaromatic group appended to the nitrogen atom were synthesized by a single-step one-pot synthesis based on the Ugi four-component condensation.
●Syntheses of peptide nucleic acid (PNA) monomers containing fluoroaromatics: Two novel classes of peptide nucleic acid monomers containing fluoroaromatics designated as F-PNA and b-F-PNA were synthesized using conventional peptide coupling methods.
●Synthesis of novel fluorophenylaryl/heteroaryl ether derivatives as selective cox-2 inhibitors: A novel class of fluorophenylaryl / heteroaryl ether derivatives were synthesized by using a variety of convenient phenolic coupling methods and subsequent hydrogenation of the coupled intermediates. The compounds synthesized are structurally similar to a number of anti-inflammatory agents.
●Synthesis of minor tautomeric isosteres of natural nucleosides as a means of studying the correlation of the tautomerization of nucleotides with DNA replication: Two minor tautomeric isosteres of natural nucleosides were synthesized where the bases used are the closest possible steric and electronic mimics of protonated enol forms of thymine and guanine (T*+ and G*+) respectively. Both the compounds were characterized as the b-anomer with the help of spectroscopic data. The 1H NMR data was matched with the data of similar known compound and the configuration was confirmed by NOE experiment.
●Enanatioselective fluorination of carbonyl compounds mediated by cinchona alkaloid derivatives/selectfluor combination: A number of carbonyl compounds were tried for enantioselective fluorination by using the combination of cinchona alkaloids/selectfluor. The fluorinated compounds were obtained in moderate yields with poor enantiomeric excess (ee).
●Total Synthesis of fluoroisosteric analogues of biologically active natural Product CJ-12662: A novel fluoroisosteric analogue of the core structure of pyrrolobenzoxazine terpenoid natural product CJ-12662 was tried to synthesize from L-Tryptophan by a combination of N-fluoro-2, 4, 6-trimethyl triflate (FTP) mediated fluorination and a Meisenheimer-type rearrangement. The product was obtained in moderate yield.
●Bioactivity guided phytochemical and pharmacological evaluation of tropical plants: The tropical having known or unknown traditional medicinal uses are studied for the presence of phytochemicals in the crude extract of the whole plant or plant parts. The crude extracts are then subjected to a preliminary bioactivity study. The biologically active extracts of fractions are evaluated for several pharmacological activities like cytotoxic, antibacterial, analgesic, anti-inflammatory, antioxidant etc. The extract or fraction is tried to purify by column chromatography or HPLC in order to isolate the compound responsible for bioactivity.
